Its best to keep putting the layers on in one go. ive done 5 layers
of copped strand , then rollered, no worries. You get a better
physical bond with the cloth that way. My glass supplier says no more
than 24 hrs between coats if adding another layer
cheers paul
Unless the project is tiny, why would you do that? The stuff sets up
really fast anyway. For any reasonable sized boat I should think the
stuff would be at least partially set before you could put the second
layer on. (Unless you're using crappy OLD resin that takes three days to
set, but that's another story.) Just be sure to use the resin that
doesn't have wax in it except for the last layer. (you can get the wax
seperately from Merton's, I think) Prepare to have your brain seared by
awful fumes, and hope the neighbors are not downwind. (I like epoxy better)

Use laminating resin and you can add layers anytime, right away or days
later. No real time critical schedule. Then use finishing resin, or add
wax to the laminating resin (the wax "floats" to the top of the resin
to seal the surface), or cover with saran wrap for the final layer
(laminating resin doesn't fully cure until oxygen deprived.)

But as Lincoln said, this is stinky stuff.... Have plenty of ventilation.

I am wondering how best to lay up multiple layers of polyester and
fiberglass quickly in one application. How soon can I put the second layer
on and would using plastic sheeting for smoothing out be ok. Would I use
the sheeting and roller to smooth each layer of glass as it is applied and
then do the next layer while the first is still wet.
